Interleukin 6 and transforming growth factor beta1 concentration in serum in patients with coronary vessels restenosis

Cytokines play a role in cardiovascular diseases by inducing inflammation, cell proliferation and apoptosis. It is well known that activated macrophages are one of the main sources of inflammatory cytokines. Previous studies have shown that the cells found in the vascular wall produce various kind of cytokines which are involved in cardiovascular diseases. It has been reported that some cytokines (IL-6, IL-8) are increased in acute coronary syndrome. The results of some other studies have suggested that PTCA induces inflammatory response in the circulation.

In the present study we investigated changes in concentration of several cytokines that are suspected to be involved in cascade mechanism after vascular injury; IL-6, a multifunctional cytokine playing a central role in inflammation and tissue injury and TGF beta which may influence atherogenesis and has a dominant protective role on coronary arteries.

We studied 36 patients with myocardial infarction who underwent stent implantation following PTCA. They underwent control coronarography after 12 months and they have been examined for lumen loss. During this follow-up the IL-6 and TGF-beta1 plasma concentration was measured in peripheral blood sample. Patients with decompensated heart failure, renal failure, hepatic failure. All patients gave informed consent. IL-6 and TGF beta concentration were measured in duplicate by ELISA using commercially available kits (R&D System). The lower limits of detection of IL-6 and TGF beta1 were 0.7 pg/ml and 7.0 pg/ml respectively. Data are expressed as mean (S.E.M.). Comparison between groups were analysed using unpaired Student’s t test. Additionally U Mann-Whitney test and Spearman rang correlation test were used. P<0.05 were considered significant. Correlation between IL-6 and lumen loss and artery stenosis were established after one year follow-up. There were significant correlation between IL-6 concentration and lumen loss and there was no significant correlation between TGF beta 1 plasma concentration and artery restenosis.
