Effect of grape procyanidins on proliferation and apoptosis of pancreas beta cells

Objective: To investigate the effect of grape procyanidins(GPC) on proliferation and apoptosis of normal pancreas beta cells and the oxygen injury cells.
Method: The cultured pancreas beta cells(BRIN-BD11) were transferred to 12-well plates. Cells were divided into two groups: oxygen injury and no oxygen injury. (1) Oxygen injury cells were damaged by 100 micromoles per litre H2O2 after pretreatment of 50 milligrams per litre,100 milligrams per litre GPC ,and (2) no oxygen injury cells were cultured with 50 milligrams per litre,100 milligrams per litre GPC only. All cells were then incubated, 37degC, 5% CO2 for 4h. Proliferation of the cells was detected by MTT assay. The apoptosis of the cells were analysized by flow cytometry. Results (1) Oxygen injury: The levels of proliferation and apoptosis of the positive control were 0.367 plus/minus 0.033 and 15.84%. The proliferation levels of 50 milligrams per litre and 100 milligrams per litre GPC groups were 15% and 32% higher, and the apoptosis levels were 5.04% and 8.81% lower than the positive control respectively. The differences were statistical(P<0.05); (2)No oxygen injury: The proliferation levels of 50 milligrams per litre,100 milligrams per litre groups and the normal control were 0.568 plus/minus 0.042 ,0.655 plus/minus 0.057 and 0.458 plus/minus 0.040 respectively. The differences were statistical (P<0.05). Conclusion GPC has protective effect on oxygen injury of pancreas beta cells. It can lessen proliferation inhibition and decrease apoptosis of pancreas beta cells induced by H2O2 .GPC can also promote proliferation of normal pancreas beta cells obviously.
